Born in Bethesda, Maryland, Sean Murray spent his childhood near Coffs Harbour in New South Wales, Australia, and holds dual U.S.-Australian citizenship. His mother Vivienne is the fourth wife of American television producer-screenwriter and NCIS series creator Donald Bellisario. It would seem nepotism comes in handy, especially in the making of a Hollywood career. Murray is not the only showbiz kid in the Bellisario clan — brother Chad W. Murray is on the production crew of NCIS and actor stepbrother Michael Bellisario and stepsister Troian Bellisario (star of Pretty Little Liars) have both appeared on NCIS.

Murray's big acting break came as Zane Grey Hart in the CBS comedy-western series Harts of the West, which featured NCIS costar Mark Harmon in a guest-starring outing as a drunken rodeo clown. Murray has appeared in several feature films, including Hocus Pocus, This Boy's Life, and Too Romantic. He joined the cast of NCIS in 2003.
